چکیده

exposure to air pollutants is significantly associated with health risks ranging from bronchial reactivity to morbidity and mortality. however, the precise mechanisms are not always fully understood. the aim of this study was to investigate the effects of sulphur dioxide (so2) on cell viability and cytokine production of a549-human pulmonary epithelial cells. test atmospheres of so2 were generated using a direct dilution method and calibrated by ion-chromatography. test atmospheres were delivered to lung cells cultured on porous membranes (0.4 μm) using harvard navicyte horizontal diffusion chamber systems. the cytotoxic endpoints were investigated using the mts (tetrazolium salt; promega), nru (neutral red uptake; sigma) and atp (adenosine triphosphate; promega) assays. expression of inflammatory markers including tumor necrosis factor-α (tnf-α) and interleukin-6 (il-6) were evaluated using double-antibody immunometric assays. dose-dependent effects of so2 were observed in a549 cells using all in vitro assays at test concentrations (10-200 ppm). the atp assay appeared to be the most sensitive test (ic50 = 48 ± 2.83 ppm) that may related to the impaired metabolic activity of the cells following so2 exposure. after analysis of tnf-α, no statistically significant differences were observed between control and exposed cells. however, the il-6 production in a549 cells was significantly reduced in a dose-dependent manner (p<0.05). these results suggest that so2 may induce a functional alteration of cells of the pulmonary epithelial preventing cells to produce adequate amounts of il-6. il-6 as a multifunctional proinflammatory cytokine may regulate cellular responses and plays a significant role in inflammation and tissue injury.

Sulphur Dioxide Exposure

Bronchoalveolar lavage (BAL) which has not previously been used in investigating the effect of sulphur dioxide (SO2) on the human lung was performed on 12 subjects before and after controlled chamber exposure with SO2 for 20 minutes. BAL fluid 24 hours after exposure with 10 mg S02/m3 (4 ppm, 10 subjects) showed increased alveolar macrophage activity as judged by an increase in lysozyme positiv...

Human bronchial epithelial cell dysfunction following in vitro exposure to nitrogen dioxide.

Nitrogen dioxide (NO2), is a major air pollutant, that causes bronchoconstriction and bronchial hyperreactivity, and may also lead to damage and inflammation of the airway epithelium. We have cultured human bronchial epithelial cells and investigated the effect of exposure to NO2, for 20 min on epithelial cell membrane integrity and function in vitro. Epithelial cell membrane damage and permeab...

Cyclic stretch attenuates effects of hyperoxia on cell proliferation and viability in human alveolar epithelial cells.

The treatment of severe lung disease often requires the use of high concentrations of oxygen coupled with the need for assisted ventilation, potentially exposing the pulmonary epithelium to both reactive oxygen species and nonphysiological cyclic stretch. Whereas prolonged hyperoxia is known to cause increased cell injury, cyclic stretch may result in either cell proliferation or injury dependi...

Mechanisms of synergistic cytokine-induced nitric oxide production in human alveolar epithelial cells.

Nitric oxide (NO) derived from inducible NO synthase (iNOS) at sites of inflammation is closely related to host defense against infection and airway inflammation. Cytokines are known to stimulate NO production in human alveolar epithelial cells in a synergistic (nonlinear or nonadditive) manner. The mechanism of this synergy is not known. We measured the activation of the transcription factor N...

Synergistic cytokine-induced nitric oxide production in human alveolar epithelial cells.

Nitric oxide (NO) is an important mediator molecule in regulating normal airway function, as well as in the pathophysiology of inflammatory airway diseases. In addition, cytokines are potent messenger molecules at sites of inflammation. The specific relationship among IL-1beta, TNF-alpha, and IFN-gamma on iNOS induction and NO synthesis in human alveolar epithelial cells has not been determined...

Rifampin augments cytokine-induced nitric oxide production in human alveolar epithelial cells.

Rifampin increased nitric oxide production and inducible nitric oxide synthase expression in alveolar cells stimulated with cytokines. Nitric oxide concentrations after induction with cytokines, cytokines with 10 microg/ml rifampin, and cytokines with 50 microg/ml rifampin were 3.2, 4.5, and 8.8 microM, respectively (P < 0.02 versus cytokines alone). This indicates that rifampin modulates the i...
